已有的流行病学研究显示,多余的脂肪组织会增加癌症的风险。而现在williamhill asia 正处于全球肥胖患病率急剧上升的时期,但是williamhill asia 对肥胖如何帮助细胞转化(非致瘤性细胞发生恶化的阶段)还知之甚少。 为了探索脂肪组织刺激转化的因素,来自密歇根州立大学的研究人员使用了一种新型的内脏脂肪组织条件培养基刺激上皮细胞在琼脂中生长的体外模型。为了扩展该系统的体内应用,研究人员使用了紫外线B诱导,内脏脂肪促进的小鼠脂质切除皮肤肿瘤模型。 研究人员发现,来自小鼠和肥胖人类的内脏脂肪能够促进非致瘤性上皮细胞在软琼脂中的生长。内脏脂肪组织的活性和成纤维细胞生长因子-2(FGF2)水平有关。人和小鼠的内脏脂肪在缺乏FGFR-1(FGF2受体)的细胞中未能刺激其在软琼脂中的生长。研究人员还证明了FGF2的循环水平与体内非黑色素肿瘤形成有关。 这些数据表明FGF2作为内脏脂肪组织释放的转化上皮细胞的主要因子,是一种全新的内脏组织促进肿瘤发生的潜在途径。消耗内脏脂肪组织中的FGF2或是在腹部肥胖个体中抑制FGFR-1,可能是重要的癌症预防策略以及改善结果的辅助治疗手段。 原始出处: D Chakraborty et al.
