2020年8月17日,中国科学院上海生化细胞所高栋,陈洛南及斯隆-凯特琳纪念癌症中心Chen Yu共同通讯在Nature Genetics 在线发表题为“Single-cell transcriptomics identifies a distinct luminal progenitor cell type in distal prostate invagination tips”的研究论文,该研究表征了来自小鼠前列腺的35129个细胞,并鉴定了以Tacstd2,Ck4和Psca表达为标志的独特的腔细胞类型(称为C型腔细胞,Luminal-C)。位于远端前列腺内陷尖端(称为Dist-Luminal-C)的Luminal-C细胞表现出更大的体外类器官形成能力和体内前列腺上皮导管再生能力。Luminal-C细胞的谱系追踪表明,Dist-Luminal-C细胞通过自我更新和分化重建了远端前列腺内腔谱系。Dist-Luminal-C细胞中Pten的缺失导致前列腺上皮内瘤变。

该研究进一步表征了11374个人的前列腺细胞,并证实了h-Luminal-C细胞的存在。该研究提供了对前列腺谱系层次的见解,将Dist-Luminal-C细胞鉴定为腔祖细胞群,并提出了前列腺癌的潜在细胞起源之一。

前列腺上皮由腔细胞,基底细胞和神经内分泌细胞组成。尽管已经报道了前列腺中存在基底干细胞和管腔干细胞,但是它们的鉴定仍然难以捉摸,并且前列腺上皮细胞谱系仍然定义不清。

使用来自分离细胞的功能性前列腺再生测定法,先前的研究表明,一些基底细胞可以产生所有三种前列腺上皮细胞谱系。最近,发现基底细胞和腔细胞均可产生含有基底细胞和腔细胞谱系的类器官。从前列腺分离并置于人工条件下的基底细胞和腔细胞具有转分化的能力。体内谱系追踪实验表明,成年小鼠前列腺的基底细胞和腔细胞都在很大程度上独立地自我维持,并且在前列腺产后发育过程中鉴定出了多能和单能祖细胞。然而,两项独立研究表明,稀有的基底细胞具有在体内前列腺再生过程中产生腔细胞的能力。

在类器官培养条件下,前列腺近端区域的Sca-1 + / Nkx3.1-腔细胞被鉴定为双能前列腺腔祖细胞。在完整前列腺的远端区域是否存在具有干细胞/祖细胞特性的腔细胞的不同亚类仍是未知的。值得注意的是,尚未确定表达在生理条件下充当前列腺腔干/祖细胞的定义的标志物的细胞类型。

前列腺癌是全球最常见的癌症之一,也是西方国家男性与癌症相关的死亡的第二大主要原因。尽管大多数人类原发性前列腺癌具有管腔表型,但是在模型系统中,基底细胞和管腔细胞均可作为前列腺癌的细胞起源。然而,尚未确定所定义的前列腺上皮细胞的干细胞样可塑性和生理条件下前列腺癌的细胞起源。最近,前列腺基底和管腔细胞群均显示出自我维持作用,并且两种细胞类型均可引发前列腺癌。然而,基底细胞的致癌转化需要基底到腔细胞的转变。此外,在某些情况下,腔内细胞显示出成为前列腺癌起源细胞的趋势更大。这些研究强调了识别腔干/祖细胞并表征其在各种情况下的行为的需要。

单个前列腺细胞的无偏特征可提供对前列腺上皮细胞谱系层次的基本了解,并指示潜在的干/祖细胞类型。最近,新兴的单细胞RNA测序(scRNA-seq)技术已允许在单个实验中查询成千上万个单个细胞中的基因表达。

在该研究中通过scRNA-seq成功地分析了35,129个成年小鼠前列腺细胞和11,374个人前列腺细胞。通过这种强大的方法,该研究定义了前列腺腔总细胞数量,并确定了在前列腺稳态和再生过程中具有腔干/祖细胞特性的候选群体。此外,该研究展示了这些细胞作为前列腺癌起始细胞起源的潜力。
