NAT COMMUN:阜外医院张浩发现肠道微生物可因缺氧促使骨髓干细胞早衰

在国家自然科学基金等资助下,中国医学科学院阜外医院张浩研究团队在干细胞研究领域取得新进展。
研究发现,慢性缺氧会导致间充质干细胞早衰,这可能是由于肠道失调引起的D-半乳糖积累。
研究将紫绀型先天性心脏病(CCHD)患者作为全身慢性缺氧的人类疾病模型,分析了慢性缺氧性患者在全身缺氧状态下间充质干细胞与肠道微生物的关系。观察到紫绀型先天性心脏病患者间充质干细胞出现早衰表型,代谢组学结果显示D-半乳糖的异常蓄积。
研究发现这些患者所经历的低氧导致肠道微生物群落多样性的改变,乳酸菌和降解D-半乳糖的酶含量显着减少。
进一步在低氧大鼠模型上,研究观察到同样的表型,并补充乳酸菌后可缓解D-半乳糖的蓄积,并挽救缺陷的间充质干细胞。
而口服补充乳酸菌可增强低氧患者间充质干细胞的功能,为系统性缺氧下患者的个性化细胞治疗提高了实验证据。
这种低氧状态不仅加剧了疾病的进程,也对各个系统器官产生一定的影响。其中肠道微生物群及肠粘膜对缺氧就极为敏感。越来越多的证据支持肠道菌群可能会影响到骨髓功能。
生理状态下,骨髓腔中处于相对的低氧状态,而这种低氧环境通常被认为是维持干细胞正常生理功能和自我更新所必需的。
然而,在全身慢性缺氧患者中,骨髓腔的低氧状态可能加重,并可能引起独特的生理变化。位于骨髓腔中的间充质干细胞以其分化为多种细胞的潜能,在许多临床试验中被应用于各种疾病的治疗中,如脊髓损伤、缺血性卒中、心肌梗死、骨关节炎等。
