Aging Cell:雷帕霉素或能有效抑制年龄相关的大脑功能退化

近日,一项刊登在国际杂志Aging Cell上的研究报告中,来自圣安东尼奥健康中心等机构的科学家们通过研究发现,药物雷帕霉素或能改善因年龄相关导致的大脑血流量(流向大脑)减少及记忆力下降的状况;相关研究结果对于开发有效抑制或治疗阿尔兹海默病的新型疗法至关重要。

近日,一项刊登在国际杂志Aging Cell上的研究报告中,来自圣安东尼奥健康中心等机构的科学家们通过研究发现,药物雷帕霉素或能改善因年龄相关导致的大脑血流量(流向大脑)减少及记忆力下降的状况;相关研究结果对于开发有效抑制或治疗阿尔兹海默病的新型疗法至关重要。

文章中,研究人员在大鼠19个月大的时候开始给其饮食中添加低剂量的雷帕霉素,日常食物中雷帕霉素疗法一直持续到大鼠34个月大(几乎是它们开始治疗时年龄的两倍),研究者发现,即使大鼠非常老了,但其大脑中血液循环的状况和开始治疗(年轻)时完全一样。这项研究中,未经治疗的老年大鼠能够反映研究者在老年人机体中所观察到的流向大脑的血液流失和记忆丧失的状况,相比之下,利用雷帕霉素治疗的老年大鼠看起来就像是中年大鼠的表现。

医学博士Sudha Seshadri表示,衰老是痴呆症的重要风险因素,这项研究中williamhill asia 意外发现,一种促进长寿的药物—雷帕霉素竟能够帮助保持老年大鼠机体大脑循环的完整性和记忆表现,目前williamhill asia 正在研究该药物对轻度认知损伤患者健康的安全性。研究人员还对未患基因工程化疾病的老年动物进行了研究,研究者表示,他们只是观察正常的衰老状况,这些大鼠存在自然的认知能力下降状况,而这并不是因强迫的疾病过程所引起的。

雷帕霉素的作用靶点—TOR是细胞生长和衰老的主要控制子,研究者发现,在机体衰老过程中,TOR能够驱动突触的缺失和大脑中血流量的减少,突触能够连接不同的神经元,而大脑依靠神经元来发送和接收信号。研究者表示,大脑会消耗大量的能量,但却没有替补资源,神经元需要葡萄糖和氧气来维持功能,血管则能够为其提供养分。最后研究者表示,后期他们还将继续深入研究来阐明雷帕霉素抑制年龄相关的大脑血管恶化的分子机制。
