前列腺癌(PCa)作为西方国家男性中最常被诊断出的恶性肿瘤之一,是癌症相关死亡的第二大主要原因。2018年,全球约有130万新确诊PCa病例。在中国,前列腺癌的发病率和死亡率逐年增加。

尽管根治性前列腺切除术或放疗被证明可以有效的控制局部PCa,但仍有一部分患者出现疾病的复发或转移。目前并没有有效的治疗方法可用于转移性PCa患者的治疗。因此,更好地了解促进PCa转移的潜在分子机制尤为重要,这也将促进新型疗法的开发以及现有治疗方案的改善实施。


MicroRNA(miRNA)是一种具有17-23个核苷酸的内源性小非编码RNA,其可以通过与靶标mRNA的3'-UTR结合而抑制转录后的基因表达。既往研究显示,miRNA能够作为肿瘤发生、侵袭和转移的各种生物过程中的重要调节因子。因此,研究miRNA在癌变和转移中的作用有助于更好地了解肿瘤的发生。

miR-199b-5p抑制肿瘤的生长和转移

据报道,miR-199b-5p在包括肝细胞癌、肾细胞癌、乳腺癌和急性髓细胞性白血病等多种癌症中表达下调,且在肿瘤细胞的生长、侵袭和转移中发挥着肿瘤抑制功能。然而,miR-199b-5p在PCa中的临床意义和作用尚不清楚。


该研究旨在通过生物信息学分析、功能丧失和获得实验以及拯救rescue实验来研究miR-199b-5p在PCa中的表达状态以及在PCa的转移中涉及的相关分子机制。


研究人员发现,与正常的前列腺组织、局部性、弱转移性和雄激素依赖性PCa细胞以及正常的前列腺上皮细胞相比,miR-199b-5p在转移性PCa组织和细胞中的表达水平显著下调。miR-199b-5p能够显著抑制PCa细胞的增殖、迁移和侵袭,且能够抑制移植瘤的生长和转移。

在PCa中DDR1是miR-199b-5p的靶标

机制研究显示,miR-199b-5p可通过直接靶向DDR1的3'-UTR区抑制该基因的表达,最终抑制由DDR1激活ERK信号通路所诱导的上皮-间质转化(EMT)相关表型。进一步研究显示,miR-199b-5p表达水平低的PCa患者的总生存期显著短于表达水平高的PCa患者,说明miR-199b-5p的缺失与PCa患者的不良预后相关。此外,在PCa中DDR1的表达水平上调,且与疾病的高Gleason分级、晚期病理分期、肿瘤的转移以及较短的总生存期显著相关。


综上,该研究结果首次揭示了miR-199b-5p在PCa侵袭和转移中的抑癌功能。该研究也确定了miR-199b-5p-DDR1-ERK这一信号通路在PCa转移中调节EMT的新机制。
