GUT:二氧化钛纳米颗粒加剧DSS诱导的结肠炎:NLRP3炎症小体起到了关键作用

西方生活方式和饮食习惯是诱发炎症性肠病(IBD)的重要因素。二氧化钛(TiO2)纳米颗粒作为一种食品添加剂广泛用于食品制作或是药物制剂中,并因此被人们大量摄入。最近,瑞士苏黎世大学的专家对二氧化钛以及核苷酸结合寡聚域受体(Nucleotide-binding Oligomerisation Domain Receptor)和热蛋白结构域中的NLRP-3炎性小体结肠炎发病过程中扮演的角色进行了相关研究。
研究人员首先用葡聚硫酸钠(DSS)成功诱导了野生型和NLRP-3缺陷型结肠炎小鼠模型,分别给予小鼠口服二氧化钛颗粒,还测试了二氧化钛对于体外培养的肠上皮细胞和巨噬细胞的促炎效果,与此同时,采用电感耦合等离子体质谱法检测了TiO2颗粒穿越肠上皮细胞屏障并在IBD患者血液中累积的能力。
实验结果显示,口服TiO2会急剧加重野生型型小鼠的结肠炎恶化,而对NLRP-3缺陷型小鼠损伤效果不明显,因此研究人员推测,NLRP-3炎性小体在结肠炎进程中起到了关键作用。同时,他们还在小鼠的脾脏中发现了大量TiO2颗粒的积累,在体外实验中,被肠上皮细胞和巨噬细胞摄取的TiO2引发了细胞内NLRP3-ASC-caspase-1的组装、caspase-1切割和NLRP-3相关的IL-1β、IL-18的释放,在UC活动期患者中,研究人员还检测到了钛水平的升高。

,如果患者的肠道屏障功能受损或是肠道本身就处于炎症状态时,很容易受到二氧化钛颗粒介导的炎症损伤,加重肠道炎症。临床医师应该建议患者减少含有二氧化钛添加剂的食品摄入。
