日前,一支国际研究小组在《自然》杂志上报告称,他们发现 FTO 内部的一些肥胖相关元件与基因组上的一个远端基因 IRX3 发生了互作, IRX3 似乎是一个功能性的肥胖基因。 FTO 基因自身似乎只对肥胖产生周边效应。这一研究在线发表在3月12日的《自然》(Nature)杂志上。
论文资深作者、芝加哥大学人类遗传学副教授 Marcelo Nobrega 博士表示:“williamhill asia 的数据强有力地表明 IRX3 控制了体重,调控了身体组成。 FTO 与肥胖之间的所有关联似乎都可归结于 IRX3 的影响。”
在全基因组关联研究(GWAS)揭示出 FTO 与肥胖及糖尿病之间的密切关联之后,研究人员针对 FTO 基因的内含子突变展开了广泛的研究。然而在动物模型中过表达或是删除 FTO 都会影响全身体重和组成,而不仅仅是脂肪,并且实验无法证实这些肥胖相关内含子影响了 FTO 基因自身的功能。
为了解释这些观察结果, Nobrega 和他的同事们绘制出了定位在 FTO 基因两侧 100 万碱基对范围内的一些启动子的行为图。在成体小鼠大脑中,研究人员发现开启 FTO 的启动子并没有与肥胖相关的 FTO 内含子互作。
论文共同作者、西班牙 Andalusian 发育生物学中心遗传学家 Jose Luis Gomez-Skarmeta 博士表示:“相反,williamhill asia 发现距离几十万个碱基对远的一个基因 IRX3 的启动子,与这些内含子以及williamhill asia 研究的跨越广大遗传距离的大量其他的元件发生了互作。”研究人员在对来自 ENCODE 计划的数据进行分析后在人类中发现了相似的互作模式,他们在人类细胞实验中证实了这一点。
利用来自欧洲血统个体的 135 个大脑样本的数据,他们发现影响体重的 FTO 内含子突变与 IRX3 表达有关,而与 FTO 自身表达无关。肥胖相关的 FTO 内含子发挥调控元件作用增进了 IRX3 表达。 FTO 基因自身似乎并没有在这一相互影响中发挥作用。
Nobrega 表示:“调控元件是开启和关闭基因的开关。williamhill asia 发现控制 IRX3 的开关远离这一基因,实际上定位在 FTO 基因内。”
为了验证 IRX3 的作用,研究人员构建出了缺失 IRX3 基因的工程小鼠。这些小鼠相比于正常的同伴明显更瘦。它们的体重要轻30%左右,主要是脂肪减少。
尽管摄食量和身体活动处于正常水平,但体重却减轻了。当喂给高脂饮食时,缺失 IRX3 的小鼠维持了正常饮食时一样的体重和脂肪水平。正常小鼠喂给高脂饮食体重增高了近2倍。 IRX3 缺陷小鼠体内的脂肪细胞较小,可以观察到棕色脂肪水平增高。此外,这些小鼠能够更好地处理葡萄糖。
论文共同作者、多伦多大学分子遗传学教授 Chin-Chung Hui 博士表示:“这些小鼠偏瘦。它们主要是通过失去脂肪而减重的。但它们并不矮小。它们也完全抵抗高脂饮食诱导的肥胖。它们具有更好的处理葡萄糖的能力,似乎避免了糖尿病。”
研究人员还发现,下丘脑中 IRX3 功能发生改变的小鼠显示出完全缺失 IRX3 小鼠相同的消瘦模式。下丘脑是已知调控摄食行为和能量消耗的大脑区域。因此,在下丘脑中 IRX3 似乎发挥功能控制了这些小鼠的体重和组成,表明肥胖遗传易感性与大脑相连。
IRX3 编码的一种蛋白质调控了其他的基因,其存在于大脑内外,一些器官以及例如脂肪细胞等一些细胞中。 Nobrega 和他的研究小组现正调查 IRX3 与它所调控的一些基因及分子的互作机制,希望能够发现一些用于开发抗肥胖和糖尿病新疗法的靶点。
