已有的研究显示,从InsP3R细胞内Ca2+释放通道到线粒体的自发Ca2+信号传导对最佳氧化磷酸化(OXPHOS)和ATP的产生至关重要。在有OXPHOS缺陷的细胞中,还原性羧化取代氧化代谢,以维持还原当量和代谢前体的数量。
为了研究线粒体Ca2+摄取在这些细胞中调节生物能量的作用,研究人员使用了OXPHOS有功能和OXPHOS缺陷的细胞。抑制InsP3R活性或线粒体Ca2+摄取增加了α-酮戊二酸(αKG)的丰度和NAD+/NADH比值,表明构成性内质网(ER)-线粒体Ca2+转移促进了αKG脱氢酶(αKGDH)的最佳活性。
减少线粒体Ca2+抑制αKGDH活性,增加NAD+,从而诱导SIRT1依赖性的自噬,该现象在OXPHOS-competent和OXPHOS-defective细胞中都存在。在OXPHOS-competent细胞中的自噬通量促进了细胞的存活,而在OXPHOS-defective细胞中,由于自噬体-lysosome融合的抑制,自噬通量受损。
在OXPHOS缺陷细胞中,αKGDH的抑制和自噬通量的受损导致细胞的死亡,以应对Ca2+从ER到线粒体的构成性通量的中断。
