肿瘤放射治疗(RT)在肿瘤病人临床治疗中占有重要的地位。在肿瘤放射治疗期间,反复低剂量的射线有可能促进肿瘤组织产生耐受性,由此导致RT疗效显着降低。放疗增敏剂可以在提高RT疗效的同时降低对正常组织的损伤,目前临床上常用的这类放疗增敏剂为硝基咪唑类或硝基苯衍生物,该类药物往往具有较大的毒副作用,其临床应用因此受到了限制。

刘鉴峰团队采用具有pH响应性多肽修饰具有放疗增敏效果的金纳米粒子,在肿瘤组织的酸性环境下形成金纳米粒子聚集体。这一策略减少了单独分散的金纳米粒子向肿瘤周边组织发生非特异性扩散和重新回到血液循环的现象,将金纳米粒子的放疗增敏值由1.16增大到1.73,从而有助于实现以较低剂量射线获得良好的放疗效果。在提高肿瘤放疗疗效的同时,由于放疗剂量降低以及正常组织内单分散的小粒径金纳米粒子可以从体内较快地清除,大大降低了射线对正常组织的损伤。该研究首次利用金纳米粒子聚集的策略实现了对肿瘤的放疗增敏,被《Advanced Science》杂志选为封底论文进行报道。
