National Science Review :突破:刘红科/赵劲/郭子建提出的新策略成功实现了肿瘤内制造靶向抗癌药物

癌症是威胁人类生命健康最严重的疾病之一,抗癌药物的研发直接关乎人的生命健康。目前临床使用的抗肿瘤药物具有效率低、选择性差、毒副作用大及耐药性等缺点。
南京师范大学刘红科和南京大学赵劲、郭子建共同通讯在National Science Review 在线发表题为“Using bioorthogonally catalyzed lethality strategy to generate mitochondria-targeting antitumor metallodrugs in vitro and in vivo ”的研究论文,该研究通过模拟合成致死的特性,选择对肿瘤及正常细胞均无毒性的钌配合物Ru-N3及化学修饰的大黄酸rhein-alkyne作为前体,利用肿瘤细胞/荷瘤小鼠肿瘤部位远高于正常细胞的铜离子浓度,通过快速高效的正交反应获得高活性的肿瘤靶向药物Ru-rhein。对照实验表明,正常细胞内不发生上述生物正交反应,故而不产生毒性。Ru-rhein靶向肿瘤细胞的线粒体(大于80%),通过自噬导致肿瘤死亡。同时建立了一种基于电感耦合等离子体质谱(ICP-MS)的新方法测定了细胞反应产物的产率。实验表明“生物正交催化致死”策略具有普适性。
总之,该策略将肿瘤细胞及活体作为抗癌药制造工厂,不仅避免了药物在运输及储藏过程中的分解和失活,也减少了药物进入体内后与生物活性分子的相互作用导致的毒副作用。“生物正交催化致死”策略为疾病精准和高效治疗的药物研发提供了新方法和平台。

为了在化学抗癌药的设计中利用合成的致死性概念,该研究开发了一种生物正交催化的致死性(BCL)策略,以在体外和体内产生靶向性抗肿瘤金属药物。通过唯一的内源性铜催化的叠氮化物炔烃环加成(CuAAC)反应,无需外部铜催化剂,即可从两种无毒物Ru-N3和大黄酸炔烃生成金属钌酸大黄素。
该研究设计了无毒物Ru-arene/Ru-N3和大黄酸炔烃来执行此策略,线粒体靶向产品Ru-rhein的生成量高(> 83%),并在体外显示出很高的抗肿瘤功效。这种BCL策略对荷瘤小鼠模型具有显著的抑癌作用。有趣的是,金属-芳烃配合物与大黄酸的结合通过CuAAC反应可以在体外和体内将两种无毒物质转化为靶向抗癌金属药物,而产物大黄酸对正常细胞则无毒。该研究阐述了大黄酸的抗癌机理,并通过增加活性氧和线粒体的损伤来诱导自噬。
该研究探讨了BCL策略的一般性,并将其扩展到其他金属络合物,例如Os-芳烃和Ir-芳烃络合物。与传统的癌症治疗方法相比,这项工作提出了一种新的方法,可通过BCL策略从基于金属的化学疗法中的无毒物体内生成靶向金属药物。
