Toxicol Sci:中科院武爱波研究组发现呕吐毒素致人源性细胞毒性相关的关键因子

国际学术期刊Toxicological Sciences在线发表了中国科学院上海生命科学研究院(营养与健康院)武爱波研究组题为“Transcription factor FOXO3a is a negative regulator of cytotoxicity of Fusarium mycotoxin in GES-1 cells”的最新研究成果,发现了呕吐毒素(DON)暴露致人黏膜上皮细胞(GES-1)线粒体毒性及其响应的关键因子FOXO3a(叉型头转录因子O亚型)。

镰刀菌毒素,尤其是呕吐毒素(脱氧雪腐镰刀菌烯醇,DON),因在各类食品中污染率、污染水平均较高,其安全限量标准的设定实际存在一些争议。如何更科学、更准确开展DON体内/体外毒理评价已成为近年来的研究热点。

博士研究生杨云霞等基于DON毒素的致呕特性,选择肠系统中的人源性细胞GES-1与DON毒素暴露互作。研究结果表明,DON暴露可诱导GES-1细胞发生氧化应激,线粒体功能失调,激活ROS-JNK-FOXO3a信号通路;活化的FOXO3a发挥转录调节作用,影响细胞的抗氧化、周期阻滞及凋亡。还通过模式生物线虫对FOXO3a(同系物DAF16)进行了体内生物学功能验证。

该研究为利用人源性细胞开展真菌毒素安全性评价工作提供了新思路。后期将进一步通过靶动物(猪、肉鸡)来探讨FOXO3a作为与DON毒性相关生物标识物的潜在可能性。

该研究得到了国家食品安全重点研发项目、国家自然科学基金面上项目等资助,同时得到了国家食品安全风险评估中心、中国农业科学院、比利时根特大学、英国利兹大学、德国明斯特大学等合作者的大力支持。
